Design Notes
The natural YcgZ promoter and the already submitted parts (BBa_K238013 and BBa_K360041) contain only the BluR operator sites around the -35 region. We designed an improved version with double the operator sites. The operator sites around the -10 region were engineered to block the polymerase from binding by overlapping the operator sequence with the -10 sequence. The -10 region was not changed in this process so polymerase binding and performance should not be impaired.
